Taking omeprazole and amitriptyline together can increase the amount of amitriptyline in your bloodstream. This may lead to an increased risk of side effects such as extreme sleepiness, dry mouth, blurred vision, constipation, or an irregular heartbeat.

Why this happens
Omeprazole can slow down the liver enzymes (specifically CYP2C19 and CYP3A4) that are responsible for breaking down amitriptyline. Because the body cannot clear the amitriptyline as quickly as usual, the levels of the medicine build up in your system.
What you should do
Back to contentsYou should use this combination with caution. If your doctor has prescribed both, they may need to adjust your dose of amitriptyline. Monitor yourself closely for increased side effects. If you feel very dizzy, confused, or notice a fast or thumping heartbeat, contact your doctor immediately.
